My opinion has always been there is much science that can be done in the temporary absence manned flight (besides Russia), although the Shuttle was important for Hubble.
The problem with expanded manned flight (moon base and mars mission) is that there are many unresolved details with few solutions in any other medium than science fiction. It was a mistake not to have a project to replace the shuttle before it was retired. This mistake was made almost a decade ago. Cameron was successful in his deep sea venture, but that required much less infrastructure than space missions. I assume they hope to use NASA facilities. American space programs will continue we just need to take a step back.
