Interesting, but perhaps we need to come up with a 289x289=83,521 or a 361x361=130,321 square sudoku. The 361x361 should have 361 19x19 subsquares. I seem to recall that even Knuth's algorithms for sudoku grow pretty fast, so simply showing that such a beast has only 1 solution may take a lot of computation. It should be not too difficult to construct a 361x361 sudoku's solution, with 19x19 subsquares. Or even bigger if you wish. And if there is -for example- only one empty cell in each subsquare, for sure there will be only 1 solution to the problem! Of course the grid will be completely stupid to fill, but I think that there is no rule asking a minimum number of empty cells in a sudoku.
